摘  要:目的研究黄芪对快速衰老鼠脑中12个衰老相关基因表达的影响。方法雄性SAM-P/8老年鼠,随机分为对照组和黄芪组,黄芪组每日腹腔注射黄芪注射液,14d后取材,RT-PCR检测。结果黄芪组与对照组比较。Gabarb3、MAPKK4和Sortilin在鼠脑额叶和海马中表达下调,CalcineurinB、MAP2和P35仅在鼠脑海马中表达下调,RAB6A在鼠脑额叶中表达下调,Calmodulin、MAP1B、RAP2A、SCN2B和Synapsin的表达在黄芪组和对照组之间比较没有统计学差异。结论黄芪可下调上述7种基因在衰老鼠脑中的表达发挥抗衰老作用,通过抑制这些基因产物对脑的消极作用抗衰老。Objective To explore the effect of Astragalus mongholicus (AM) on the expression of twelve agingassociated genes in the brains of SAM-P/8 mice. Methods Male senescence accelerated mouse/prone 8 ( SAM-P/8 ) mice were divided into control group and AM group. RT-PCR was used to detect the changes of the expressions of twelve aging-associated genes in frontal lobes and hippocampus 14d after injection of AM into abdominal cavity in AM group. Results Gabarb3 and MAPKK4 and Sortilin showed reduced expressions in frontal lobes and hippocampus, calcineurinB and MAP2 and P35 decreased in hippocampus, and RAB6A decreased in frontal lobes in AM compared with control.There were no significant differences for calmodulin, MAP1B, RAP2A, SCN2B and Synapsin in AM compared with control(P 〉 0.05). Conclusion Astragalus mongholicus can downregulate the expressions of above seven aging-associatd genes in the brains of SAM-P/8 mice to have antisenility effect through inhibition of these gene products.
