摘 要:目的观察复方天麻蜜环菌糖肽片对大鼠脑缺血再灌注后大脑皮质缺血灶周围区caspase-3蛋白、凋亡诱导因子(AIF)时相表达及对神经细胞凋亡的影响。方法应用线栓法建立Wistar大鼠大脑中动脉阻塞(MCAO)再灌注模型,氯化三苯基四氮唑(TTC)染色观察梗死灶的分布,应用原位末端标记(TUNEL)和免疫组化技术分别观察模型组、复方天麻蜜环菌糖肽片治疗组和对照组(生理盐水干预组)大脑中动脉栓塞90min再灌注6h、24h、72h时缺血灶周围脑区caspase-3蛋白、凋亡诱导因子和TUNEL阳性细胞的时相表达。结果脑缺血再灌注后在缺血灶周围区均出现三者的表达,且表达有几乎一致的规律性。栓塞90min再灌注24h、72h,复方天麻蜜环菌糖肽片治疗组与其他两组比较,caspase-3、凋亡诱导因子阳性细胞和TUNEL染色阳性细胞的数量均明显减少(P〈O.01)。结论AIF、caspase-3参与了局灶性脑缺血后神经细胞的凋亡过程,复方天麻蜜环菌糖肽片可以减少AIF、caspase-3蛋白的表达,减轻迟发性神经元死亡,对缺血性脑组织有保护作用。Objective To investigate the effect of compound armillaria mellea on the expressions of apoptosis-inducing factor(AIF), caspase-3 protein and neuron apoptosis in perifocal areas after ischemia-reperfusion in rats. Methods Middle cerebral artery occlusion(MCAO) and reperfusion in the rats were used as the research model. Infarction zones were confirmed by 2, 3, 5-triphenyl tetrazolium chloride (TTC) staining. The expressions of AIF,caspase-3 positive cells and variations of neuron apoptosis in perifocal regions in model groups injured by ischemia for 90 minutes, then reperfusion at 6, 24, 72 hour points, treatment groups injected with compound armillaria mellea and control groups(injected with NS) were studied using immunohistochemistry and in situ apoptotic detection ( Tunel method). Results The expressions of AIF, caspase-3 and Tunel positive neurons had the similar regularity in perifocal areas at different time points after ischemia-reperfusion. The numbers of AIF, caspase-3 and Tunel positive neurons in groups injected with compound armillaria mellea were significantly decreased compared with those in model groups and in control groups at 24, 72 hour points in perifocal areas after ischemia-reperfusion. Conclusions AIF and caspase-3 may participate in the neuron apoptotic process after focal cerebral ischemia and reperfusion. Compound armillaria mellea may have a protective function for the cerebral tissues after ischemia-reperfusion by reducing the expressions of AIF,caspase-3 and neuron apoptosis.
