摘 要:目的检测不同胎龄人胚原始生殖细胞中Oct-4、人端粒酶逆转录酶(hTERT)的表达变化。方法取人胚胎生殖嵴,通过RT-PCR检测5-13周龄人胚生殖嵴中Oct-4、hTERT的表达变化;同时,取人胚胎生殖嵴,经石蜡切片,HE染色、免疫组织化学染色等技术,观察不同胎龄人胚原始生殖细胞的形态,检测Oct-4、hTERT的表达情况。结果胎龄6-7周时,生殖嵴中可见少量原始生殖细胞,且表达Oct-4及hTERT;胎龄8~12周时,生殖嵴中原始生殖细胞数量增多,Oct-4及hTERT表达增强(P〈0.05);胎龄13周以后,生殖嵴中Oct-4阳性的原始生殖细胞数量逐渐减少,Oct-4表达减弱,但hTERT仍然高表达。结论不同胎龄人胚原始生殖细胞中Oct-4的表达呈动态变化,胎龄8-12周时表达较强,但hTERT的表达量始终维持在较高水平,没有明显变化。Objective To analyse the expression levels of Oct-4 and human telomerase reverse transeriptase(hTERT) in human primordial germ cells (PGCs) which were isolated at different fetus ages. Methods The expressions of both Oct-4 and hTERT were detected by RT-PCR. The gonadal ridges of human embryos were embedded with paraffine and sliced, and stained with HE. The morphology of the human primordial germ cells was observed. The gonadal ridges were analyzed for the expression of Oct-4 and hTERT through immunohistochemistry. Results There were a few PGCs in the gonadal ridges in the sixth to the seventh week of gestation, and both Oct-4 and hTERT were expressed in the PGCs. The PGCs with Oct-4 and hTERT positive staining increased in number in week 8 to 12 of gestation (P 〈 0.05 ). The PC, Cs decreased in number and the Oct-4 immunostaining intensity in PGCs declined, but the hTERT was still expressed in PGCs in the thirteenth week of gestation. Conclusion The immunostaining intensity of Oct-4 varies with different stages of development and is the highest during week 8 through 12. There was no obviously change on the expression of hTERT.
