摘 要:目的探讨氟伐他汀对粥样硬化家兔CD68和增殖细胞核抗原(PCNA)表达的影响。方法采用高脂饮食喂养法建立动脉粥样硬化(AS)模型。将24只家兔随机分为:正常对照组、AS模型组和氟伐他汀组,每组8只。饲养12周后处死,取颈动脉行HE染色,免疫组化染色法检测PCNA和CD68的抗原表达。结果氟伐他汀组血清总胆固醇(TC)、低密度脂蛋白-胆固醇(LDL-C)浓度明显低于AS模型组(P<0.01)。AS模型组颈动脉斑块中见大量CD68及PCNA阳性细胞,氟伐他汀组对应阳性细胞数显著少于AS模型组(P<0.01)。结论氟伐他汀治疗可以抑制高胆固醇血症兔颈动脉粥样斑块的形成,减轻斑块内巨噬细胞的浸润并抑制PCNA的产生,从而起到稳定斑块的作用。Objective To observe the expressions of CD68 and proliferating cell nuclear antigen( PC- NA) in carotid atherosclerotic plaques in hypercholesterolemic rabbits and the effects of fluvastatin on them. Methods The carotid atherosclerosis model of rabbit was established through the inducement of high cholesterol. 24 rabbits were randomly divided into 3 groups: control, hyper-lipid diet and Fluvastatin( n = 8 in each group). The levels of TC, TG, LDL-C in Serum were measured at different time points. These rabbits were killed after 12 weeks. Carotid arteries were isolated and paraffin-embedded slices were performed. Immunohistochemical staining of CD68, PCNA expression were performed with S-P method. Results Serum concentration of TC and LDL-C in fluvastatin group was lower than in hyper-lipid diet group (P 〈 0.01). The number of CD68 and PCNA positive cells of carotid plaques in hyper-lipid diet group was more than that in fluvastatin group( P 〈0.01). Conclusion Fluvastatin could prevent the formation of carotid atherosclerotic plaques in hypercholesterolemic rabbits. Fluvastatin could stabilize carotid plaques through decrease of macrophages infiltration and inhibit the expression of PCNA.
