摘  要:观察Ti-Nb-Zr-Sn系钛合金两种表面形态对兔腱膜成纤维细胞生物学行为的影响,评价其生物相容性。采用组织块法培养原代兔跟腱腱膜成纤维细胞并传代后,接种至光滑表面和规则微纹理表面两组钛合金薄片上,通过MTT比色法、扫描电镜等技术,观察细胞在不同微形态钛合金表面的增殖和贴附状况。兔腱膜成纤维细胞在两种形态钛材料表面上的细胞增殖速率不同,其增殖速率的吸光度峰值在光滑组为0.228,微纹理组为0.316,有明显统计学差异(P<0.01)。扫描电镜下观察,光滑组成纤维细胞扁平,伪足少,贴附无定向性;规则微纹理组的细胞胞体饱满、伪足数量较多且相互连接,沿材料表面纹理贴附,其长轴与纹理方向一致。Ti-Nb-Zr-Sn系钛合金的规则微纹理表面处理有利于动物成纤维细胞的附着,其生物相容性优于光滑表面。[Objective] To observe surface topographic effects of Ti-Nb-Zr-Sn alloy on rabbit lacertus fibroblastic cell behaviors and evaluate its biocompatibility in vitro. [Methods] Cultured by methods of explant and passaged, fibroblasts dissected from rabbit achilles' tendon were inoculated on two groups of titanium alloy slices with smooth and microtextured surface. Their proliferation and adheresion ability were examined by MTT test and scanning electron microscope (SEM). [Results] The fibroblasts had different proliferation ability on the two surfaces, the peak absorbance on smooth group was 0. 228 and the microtextured group was 0. 316. The results were statistical significance. (P〈0.01). By the observation of SEM, astatism flat cells with few pseudopods attached on smooth surace, while satiate cells with a great deal of interconnected pseudopods attached on microtextured surface, cells oriented along, the surface texture. [Conclusion] The microtextured surface of Ti-Nb-Zr-Sn alloy is good for the adhesion of fibroblasts and its biocompatibility is superior to the smooth surface.
