巴西橡胶树HbCOI1基因5′调控序列的克隆及分析

摘　　要：利用GenomeWalker方法获得了巴西橡胶树HbCOI1的5’调控序列。通过分析表明，在该序列中除了含有真核生物典型的核心启动子区域（879-928bp，TATAbox存在于886bp）外，一些与真核生物顺式调控元件相似的序列也存在其中，如在63、85和604bp等处具有CAAT序列；在212、385和427bp处有5’UTRPy-riehstretch元件；在96bp处有1个与茉莉酸应答相关的元件TGACG；在43bp处存在1个与水杨酸应答相关的TCA—element；在145bp处存在1个与抗性和胁迫应答相关的TC丰富重复序列。此外，在626、745、834和864bp处分别有ACGTT—box、GATA—box、ARRlAT和bbox等顺式作用元件。The 5＇ regulatory region of HbCOI1 from Hevea brasiliensis was isolated using GenomeWalker. TATA box and other core configurations were found in promoter region. Several sequences similar to eukaryotic cis-regulatory element were found in the 5＇-UTR proximal 5＇ flanking sequence of HbCOI1. At position 63 bp,85 bp and 604 bp,CAAT sequences could be found,which is similar to the CCAAT motif found in many higher eukaryotic promoters. The 5＇ UTR Py-rich Stretch was found at 212 bp,385 bp and 427 bp. The cis-acting regulatory elements involved in the MeJA-responsiveness and abscisic acid responsiveness were found at 96 bp and 43 bp. In addition, ACGTT-box,GATA-box, ARR1AT and I-box were presented at many positions,which is similar to the other plant promoter consensus elements.
