南方鲇细胞色素b基因序列的保守性

摘  要:对5个种群的南方鲇(Silurus merdionalisChen)的线粒体DNA细胞色素b基因片段的500 bp序列进行测定,经Clustal X同源排序后得400 bp序列,结果发现:南方鲇种群内碱基的变异很低,雅砻江、岷江、乌江种群均为0,宜宾种群为0.25%,洞庭湖种群为1%;雅砻江、岷江、乌江种群只有1种单倍型,宜宾种群有2种单倍型,洞庭湖种群有5种单倍型,且单倍型间变异位点很少,雅砻江、岷江、乌江种群无变异位点,宜宾种群仅有1个变异位点,洞庭湖种群有4个变异位点.表明细胞色素b基因在南方鲇种群内是相当保守的.500 bp fragments of cytochrome b gene were obtained by PCR method from the mitochondrial DNA of Silurus merdionalis from five populations (four populations from the upper reaches of the Yangtze River (the Yalongjiang River, the Minjiang River, the Wujiang River and around Yibin, Sichuan) and a population from the middle reaches of the Yangtze River (Dongting Lake)), and a mitochondrial cyto-chrome b DNA of 400 bp sequence was successfully sequenced by Clustal X homologous sequence analysis.Base variation rate in the five populations was found to be very low, being 0 in the Yalongjiang River, the Minjiang River and the Wujiang River populations, 0.25% in Yibin population and 1% in Dongting Lake population. There were five haplotypes in Dongting Lake population, two in Yibin population and one in Yalongjiang, Minjiang and Wujiang populations. Variable sites among haplotypes were few. No variable site was noticed in Yalongjiang population, Minjiang population and Wujiang population, one was noticed in Yibin population and four Dongting Lake population. These results indicated that cytochrome b gene is very conservative in the five populations of S. merdionalis.
