摘  要:目的研究大果山楂黄酮提取物对大鼠实验性慢性肝损伤、肝纤维化的预防作用,并探讨其作用机制。方法采用40%CCl4皮下注射制备肝纤维化大鼠模型,分别用3种不同浓度的大果山楂黄酮提取物进行干预,观察检测大鼠血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、血清白蛋白与血清球蛋白的比值、肝组织匀浆超氧化物歧化酶(SOD)和丙二醛(MDA)含量以及肝组织病理学变化。结果大果山楂黄酮提取物预防给药能降低CCl4所致大鼠血清中异常增高的ALT,AST;升高CCl4所致大鼠血清中异常降低的A/G比值;大果山楂黄酮提取物预防给药能使肝损伤大鼠肝匀浆异常降低的SOD含量上升,异常增高的MDA含量下降;光镜下观察肝纤维化模型组大鼠肝细胞严重变性、坏死,胶原纤维明显增加,大果山楂黄酮提取物预防各组大鼠肝细胞病理损伤得到明显改善。结论大果山楂黄酮提取物对大鼠实验性肝损伤具有一定程度的预防作用和抗肝纤维化作用。Objective To study the anti -hepatofibrotic effect of the Malus doumeri(Bois)Chev, flavonoids extraction on experimental hepatic fibrosis and its mechanism. Methods The hepatic fibrosis rat model was developed by subcutaneous injection of 40% CCl4 (5 ml/kg) and rats were treated with extract of Mdf at 1.5g/( kg·d), 0.75/( kg·d) or 0. 38/( kg·d). Alanine aminotransferase ( ALT), aspartate aminotransferase ( AST), Albumin ( A ), globulin ( G ) levels were measured and pathological changes after of liver tissue was examined HE staining. Results The extract of Mdf could reduce the serums levels of ALT and AST, increase the hepatic ratio of A/G, increase the activity of superoxide dismutase and reduce the level malondialdehyde Examined by light microscope, the degeneration and necrosis level of hepatocytes and the distribution of collagen fibrosis were markedly increased in the model group. The extract of Molf could significantly alleviate pathological changes and the collagen fibrosis. Conclusion The extract of Mdf can protect hepatocytes from injury and prevent the progression of hepatic fibrosis in rats.
