摘  要:目的探讨宫内缺氧对新生大鼠神经干细胞增殖的影响及当归注射液的保护作用。方法将孕期14 d的SD大鼠随机分为对照组、缺氧组和当归治疗组。缺氧组孕鼠自孕第14天开始每天一次经尾静脉注射生理盐水8 m l/kg(连续7d),后用低张性缺氧模型致胚鼠宫内缺氧2 h(连续3d:孕期第14,15,16天);当归治疗组用250 g/L的当归注射液代替生理盐水,其余处理同缺氧组;对照组不缺氧,其余同缺氧组。孕鼠分娩后立即取新生鼠大脑组织,常规石蜡切片,经巢蛋白(nestin)免疫组织化学染色后行图像分析。结果缺氧组新生鼠海马巢蛋白免疫组化阳性细胞的积分光密度(IOD)值较对照组增加[IOD缺氧组:(14 806±4 636.8);IOD对照组:(7 387±3191.7),P<0.05];当归治疗组新生鼠海马巢蛋白免疫组化阳性细胞的IOD值较缺氧组减弱[IOD治疗组:(9 859±2 386.4),P<0.05]。结论一定程度的缺氧可刺激神经干细胞增殖;当归注射液对宫内缺氧新生大鼠神经干细胞可能具有保护作用。Objective To study the effect of i ntrauterin hypoxia on the proliferation of neural stem cells(NSCs) from newborn rats and the protective role of Angelica injection on NSCs in a state of hypoxia. Methods 14 d - conceiving female SD rats were divided randomly into three groups:control group(CG) , hypoxia group (HG) and Angelica group(AG) with five rats in each group. Since the fourteenth day of pregnant the rats of HG were injected with Normal Saline through caudal veins (within 7 straight days ; 8ml/kg. d)before intrauterin hypotonic hypoxia (within 3 straight days;on the fourteenth, fifteenth and sixteenth day after pregnancy;2 hours everyday ). Angelica sinensis injection instead of Normal Saline was injected into the rats in the AG ,and other procedures were the same as the HG. No administration of hypoxia was performed in the CG. Their brain tissues of newbom ruts were hamested immediately of newborn rats after childbirth. The expression of nestin in the hippocampus of newborn rats was studied with immunohistochemistry and the results were analyzed by image processing system. Results IOD of nestin positive cells in HG was higher than that of the CG( IODHG : 14806 ± 4636.8 ; IODCG :7387 ± 3191.7, P 〈 0.05 ) ; but in AG,it was lower than that of HG(IODAG :9859±2386.4 ,P 〈0. 05 ). Conclusion Hypoxia can stimulate the proliferation of the NSCs Angelica injection can play a protective role on NSCs from newborn rats with intrauterin hypoxia.
