摘　　要：目的研究白细胞介素-1β(IL-1β)对椎间盘髓核中蛋白多糖代谢的影响。方法取自然流产的胎儿一具,术后4h内无菌取出椎间盘,进行髓核组织的体外培养,培养液中分别加入0.1nmol/L、0.2nmol/L、0.4nmol/L的IL-1β,30h后,检测培养液中硫酸软骨素的含量。在髓核组织培养液中加入0.3nmol/L的IL-1β,分别培养12h、24h、72h后,检测硫酸软骨素的含量。结果①IL-1β实验组较对照组培养液中硫酸软骨素的含量增加,并且随着IL-1β浓度的增高。②随着时间的增加,培养液中硫酸软骨素的含量增加。结论IL-1β对椎间盘基质中蛋白多糖有降解作用,并呈现出浓度和时间依赖性。Objective To study the effects of interleukin-1β（IL-1β）on the metabolism of proteoglycan in the nucleus pulposus of lumbar intervertebral disc.Methods A fetus specimen obtained from spontaneous abortion was used for this study.The lumbar intervertebral disc was removed and cultured in vitro.The content of chondroitin sulfate in the culture medium was measured by Alcian method in 30 hours after the addition of IL-1β（0.1,0.2 and 0.4 nmol/L respectively）,and in 12,24 and 72 hours after the addition of 0.3 nmol/L of IL-1βto the tissue culture medium of the nucleus pulposus.Results ①The concentration of chondroitin sulfate with the addition of IL-1β increased than that in the controls.②The content of the chondroitin sulfate increased progressively along with the time of culture.Conclusion IL-1βcan stimulate degradation of proteoglycan of the lumbar intervertebral disc.The effect shows concentration and time dependent.
