摘  要:目的:研究中药蛇葡萄根提取物的含药血清对HSC/T6凋亡及凋亡基因Bax/Bcl-2表达的影响,从细胞学和分子生物学水平探讨中药蛇葡萄根抗纤维化的作用机制。方法:用流式细胞仪测定各体积分数(5%,10%和20%)的含药血清对HSC/T6作用后的DNA含量,计算细胞凋亡率;用SABC染色检测凋亡基因Bax和Bcl-2的蛋白表达。结果:含药血清体积分数为5%,10%和20%的蛇葡萄根提取物对HSC/T6的凋亡率分别为8.50%,14.30%和22.65%,均显著大于含生理氯化钠溶液的血清对照组(P<0.01);且均上调HSC/T6的促凋亡基因Bax的蛋白表达,同时下调抗凋亡基因Bcl-2的基因表达。结论:蛇葡萄根提取物含药血清能诱导HSC/T6的凋亡,上调促凋亡基因Bax的表达,下调抗凋亡基因Bcl-2的表达。Objective: To investigate the effect of Radix Ampelopsis Sincae extract-containing rat serum on the apoptosis and expression of Bax/Bcl-2 gene in cultured HSC/T6 cells, the hepatic stellate cell line, and its possible antifibrotic mechanism at cellular and molecular levels. Methods:Flow cytometry was used to detect the contents of DNA in HSC/T6 cells, and SABC was used to detect the expression of Bax/Bcl-2 gene. Results: The apoptotic rates were 8.50% , 14.30% and 22.65% after treatment with 5% , 10% and 20% Radix Ampelopsis Sincae extract-containing rat serum, which were more than those treated with control serum (P 〈 0.01 ). The gene expression of Bax was up-regulated and that of Bcl-2 was down-regulated after treatment with the extract-containing rat serum. Conclusion:Radix Ampelopsis Sincae extract-containing rat serum induces apoptosis, up-regulates the expression of Bax gene and down-regulates that of Bcl-2 gene in HSC/T6 cells.
