摘 要:目的:研究P2X7受体介导的小胶质细胞形态和受体表达的变化,探讨P2X7受体信号通路及其下游过程。方法:运用ATP刺激原代小胶质细胞,免疫细胞化学观察P2X7受体介导的细胞形态和受体表达的变化;建立表达P2X7受体的单克隆细胞模型,免疫细胞化学观察ATP刺激下P2X7受体介导的细胞形态和受体表达的变化。结果:ATP刺激下小胶质细胞和P2X7-HEK293细胞的突起变短,形态变圆,体积缩小;随着ATP刺激量的增加,P2X7受体的表达逐渐增加。结论:P2X7受体介导了小胶质细胞形态的变化。Objective: To study the morphological changes mediated by P2X7 receptors and its expression in microglial cells, and explore its signaling and downstream processing. Methods: After primary microglial cells and monoclonal HEK293 cell transfected by P2X7 receptor stimulated by ATP, the morphological changes mediated by P2X7 receptor and its expression were observed with immunocytochemistry in these cells. Results: The processes of the primary microglial cells and P2XT- HEK293 cells disappeared; their shapes changed from irregular to round and their volumes became smaller after ATP stimulation; and the level of P2X7 receptor expression increased gradually with increasing of the concentration of ATP. Conclusion: P2X7 receptors mediate the morphological change in microglial cells.
