摘  要:目的:探讨部分去背根及部分去背根后针刺对备用背根节神经元血小板源性生长因子(PDGF)表达的影响。方法:制备猫双侧备用背根节模型,并针刺位于右侧备用根支配区的两组穴位,针刺7d及14d后分别取针刺、非针刺侧备用背根节,行免疫组织化学ABC法显色。分别计数各组阳性神经元总数以及大、中小阳性神经元的数量。结果:PDGF在备用背根节的大、中小神经元均有表达。部分背根切断后,针刺侧、非针刺侧PDGF阳性神经元总数及中小阳性神经元数在7d时较正常显著减少,针刺后7d时,PDGF阳性总数及大神经元数针刺多于非针刺侧,针刺14d时,阳性总数及中小神经元数多于非针刺侧。结论:部分背根切断及针刺可影响PDGF在备用背根节的表达,这种作用显效的时间因不同时相、不同类型的背根节神经元而异。Objective: To investigate the expression of platelet derived growth factor (PDGF) in spared dorsal root ganglion after partial dorsal root rhizotomy and the application of acupuncture. Methods: Fifteen adult cats were divided into three groups. Five cats were kept intactly in the control group. Ten cats were subjected to bilateral partial dorsal root rhizotomy (L1-L5, L7-S2 DRGs were sectioned, and L6 DRG was spared). On the right leg of these operating groups, two sets of acupoints (Zusanli and Xuanzhong; Futu and Sanyinjiao) located in the distribution area of spinal nerve L6 were electro-stimulated alternatively 30 min everyday by electro-needling. The left leg was not treated with acupuncture. On the 7th and 14th day after partial dorsal root rhizotomy and acupuncture, the animals in these groups and the control group were sacrificed. The spared DRGs (L6) were taken, made into frozen sections and then stained by immunohistochemistry. The positive large, medium-small sized neurons in spared DRGs were counted. Results: PDGF-immunoreactivity was distributed in large and medium-small sized neurons. The total and medium-small sized PDGF positive neurons were found to decrease apparently on the 7th day. The number of positive total and large neurons of PDGF on the acupuncture side increased apparently compared with those on the non-acupuncture side at 7th day. On the 14th day, the positive total and medium-small sized neurons increased apparently compared with those on the non-acupuncture side. Conclusion: Partial dorsal root rhizotomy and acupuncture could affect the expression of PDGF in spared DRG and this function have influence on different type of DRG neurons in different periods.
