摘 要:绵羊(Ovis arise)卵母细胞经丁内酯-I(BL-I)抑制培养,然后转入成熟培养液培养不同时间,进行体外受精并观察胚胎发育情况。结果表明,解除8h抑制再培养8h组处于MⅡ期卵母细胞率6.38%,显著低于其它组(P<0.01)。继续培养16和20h的卵母细胞成熟率与对照组(常规培养24h)差异不显著,培养24h组卵母细胞成熟率(70.83%)稍高于对照组(66.18%),但差异不显著(P>0.05)。培养16和20h组囊胚率分别为2.99%和13.64%与对照组24.39%差异显著(P<0.05),培养24h组的囊胚率20.33%与对照组差异不显著(P>0.05)。表明BL-I对绵羊卵母细胞的体外成熟有明显抑制作用,但其抑制作用是可逆的,而经BL-I处理后未能提高绵羊卵母细胞体外受精和胚胎的整体发育能力。In order to investigate nuclear maturation , ovine (ovis arise) oocytes were in vitro cultured by inhibiting with butyrolactone Ⅰ (BL- Ⅰ ). They were moved, after 8 h, and in vitro matured (ⅣM) randomly in appropriate medium for different durations to analyze the synchronization of both their nuclear and cytoplasmic maturation. The results showed that only the rate of ovine oocytes in M Ⅱ stage, in ⅣM group for 8 h, was significantly lower than that of the others (P 〈 0.01), but there were no significant differences of both oocyte maturation and blastocyst rates among all the groups (P 〉 0.05). It suggests that such kind of inhibition by BL-Ⅰ should be reversible although, it may not improve the cleavage and developmental competence ofovine embryos.
