TNFα-308G/A和TNFβG252A基因多态性与阿茨海默病的关系

摘　　要：目的探讨肿瘤坏死因子TNFα-308G/A和TNFβG252A基因多态性与阿茨海默病(AD)的关系。方法用酶免法测定TNF水平,运用多聚酶链反应技术检测66例阿茨海默病及143例正常人肿瘤坏死因子TNFα-308G/A和TNFβG252A基因多态性。结果AD组肿瘤坏死因子TNFα-308G/A1/1和TNFα-308G/A1/2,TNFα-308G/A2/2表型频率分别为:0.8636,0.1212,0.0152,对照组分别为:0.8881,0.1049,0.070;AD和TNFα-308G/A1,TNFα-308G/A2基因频率分别为:0.9242,0.0758,对照组分别为:0.9021,0.0979;AD组肿瘤坏死因子TNFβG252A1/1,1/2,2/2表现频率分别为:0.242,0.591,0.167,对照组分别为,0.252、0.573、0.175;AD组TNFβG252A1,TNFβG252A2基因频率分别为:0.5379,0.4621,对照组分别为:0.5385、0.4615。肿瘤坏死因子TNFα-308G/A和TNFβG252A各种基因型频率在患者组与正常对照组之间的差异无显著性(P>0.05),而血浆中TNF水平,AD组明显高于对照组,两组之间比较差异有显著性(P<0.05)。结论血清TNF水平显著升高,提示炎性反应在AD病程中有重要作用,肿瘤坏死因子TNFα-308G/A和TNFβG252A基因多态性与AD无明显相关。Objective To investigate the relationship between plasma levels of tumor necrosis factor（TNF） or its polymorphism and Alzheimer＇ s disease. Methods Plasma level of TNF was measured by RIA and the polymorphism of TNFα-308G/A and TNFβG252A was screened by PCR - RELP. Results The gene frequency of TNFα-308G/A in AD group was as following： and TNFα-308G/A 1/1,1/2 and 2/2 genotype frequency： 0. 8636,0, 1212 and 0. 0152, TNFα- 308G/A 1 and TNFα- 308G/A 2 Genefrequency ： 0. 9242 and 0. 0758 o the gene frequency of TNFα- 308G/A in control group was as followed; TNFα- 308G/A 1/1,1/2,2/2 genotype frequency： 0. 8881,0. 1049 and 0. 070, TNFα- 308G/A 1 ,TNFα-308G/A 2 Genefrequency： 0. 9021 and 0. 0979, The gene frequency of TNFβG252A in AD group was as following; TNFβG252A 1/1, 1/2 and 2/2 genotype frequency： 0. 242, 0. 591 and 0. 167, TNFβG252A 1 and TNFβG252A 2 gene frequency ： 0. 252,0. 573 and 0. 175. The gene frequency of TNFβG252A in control group was as following; TNFβG252A 1/1,1/2 and 2/2 genotype frequency： 0. 252,0. 573 and 0. 175, TNFβG252A1 and TNFβG252A 2 Genefrequency： 0. 9021 and 0. 0979. There were no significant differences in the frequencies of TNFα-308G/A and TNFβG252A gene between the patients and control group （ P 〉 0. 05 ）. Alzheimer＇ s disease group showed significantly higher plasma level of TNF than the control group （ P 〈 0. 05 ）. Conclusion Plasma TNF increases in Alzheimer＇ s disease. The results suggested： TNF may play important role in the procession of AD and no evident association was found between TNFα- 308G/A or TNFβG252A polymorphism and Alzheimer＇ s disease.
