牛布鲁菌单克隆抗体乳胶凝集试验方法的建立

摘　　要：建立了用纯化好的牛布鲁菌(B.abortus)单克隆抗体致敏乳胶的检测方法。通过试验确定了抗体致敏乳胶最佳偶联蛋白量为1.23 mg/mL,最佳致敏时间4 h,最佳的乳胶浓度为1%。水样模拟样本的制作,取含1.0×109cfu/mL灭活的B.abortus544A生理盐水菌悬液各1 mL,加入9 mL自来水中,充分混合均匀,各取1 mL,土样、奶样模拟样本制作同上。最低检出率水样为3×104cfu/mL^1.0×105cfu/mL,土样、奶样为5×104cfu/mL^1.0×105cfu/mL。The purified B. abortus monoclonal antibodies was used to sensitize latex. The most suitable condition for coating the latex was determined by square matrix titration. The results showed that the optimal concentration of the coated monoclonal antibody was 1.23 mg/mL, the optimal time of the sensitized latex was 4 h,the optimal latex concention was 1%. Preparation of simulative samples（the water samples）, 1 mL of inactivated B. abortus 544A that concentration is 1.0 10^9 cfu/mL in physiological saline,was mixed with tap water 9 mL, thorough mixing. Preparations of milm samples and oil samples were the same as mentioned above. The lowest detection rate in water samles was 3 × 10^4-1.0 × 10^5 cfu/mL, and were 5× 10^4 -1.0×10^5 cfu/mL in il samples and milm samples.
