摘 要:目的采用基因芯片方法,筛查低水平铅暴露对仔鼠海马基因表达的影响。方法建立出生前后低水平铅暴露大鼠的动物模型,即妊娠期和哺乳期给予母鼠0.2%醋酸铅溶液自由饮水,仔鼠断乳时测定其血铅水平,鉴定模型成功后提取仔鼠海马总RNA,采用[α-33P]dATP标记的基因芯片检测铅对仔鼠海马基因表达谱的影响。结果实验组血铅水平(3.41±0.27)μmol/L高于对照组(0.29±0.06)μmol/L,P<0.05。基因芯片筛查发现有130个基因出现2倍以上的表达变异,其中120个上调,10个下调,多为已知功能基因,上调的包括跨膜信号传递系统的系列基因、神经代谢和发生相关的基因、神经传导相关的基因及炎症因子基因等,下调的包括与凋亡和细胞应激相关的基因。结论出生前后低水平铅暴露引起断乳期大鼠海马基因表达谱的变化,可能是发育期铅暴露影响学习记忆等海马主要生理功能的物质基础。Objective To investigate the impact of lead exposure during the pre-and post-natal period on gene expression of hippocampus in developmental rats in vivo with cDNA microarray.Methods Pregnant Sprague-Dawley rats at(18±0.5)days of gestation were administered lead acetate(0.2%)in their drinking water.Dosing began 3 days prior to breeding and continued until their pups were weaned at 21 days.At the time of weaning,blood lead level of pups were examined and hippocampus of pups were dissected.Total RNA was isolated and treated according to specific instructions.Nylon membrane was used as the array base.Oligonucleotide probes were synthesized and spotted onto the nylon membrane to explore the variation of gene expression.Results Lead-exposed rats had higher blood lead level than controls[(3.41±0.27)μmol/L vs(0.29±0.06)μmol/L,P〈0.05].120 up-regulated and 10 down-regulated genes were identified by cDNA microarray,Pb/C score greater than 2 or less than 0.5 were considered the significantly altered genes after specific treatment in the study.Conclusion Lead exposure during late gestation and lactation period may influence learning and memory of rat due to changes of some differentially expressed genes.
