摘　　要：中国首例人类免疫缺陷病毒（ＨＩＶ－１）Ａ亚型毒株的鉴定苏玲邢辉羊海涛１罗小光２邵一鸣（中国预防医学科学院艾滋病参比实验室，北京１０００５０）关键词人类免疫缺陷病毒（ＨＩＶ），艾滋病（ＡＩＤＳ），亚型，套式聚合酶链式反应（ｎｅｓｔｅｄ－ＰＣＲ），序列分...At the beginning of the 1990s,HIV/AIDS has quickly emerged to be a public healthth reaten to Asia.The presence of several subtypes of HIV-1 in several areas of Asia suggests multiple introductions of different HIV subtypes.A rapid increase of HIV infection was clearly seen in many provinces of China since 1994.Apart from Yunnan,other epidemic areas were found with subtypes B,B’,C and E in China.The identification and characterization of the first subtype A HIV-1 in China was reported here.The subtype A HIV-1 is known commonly in A frica and later found in India.We got the samples from people returned after working in Africa.The mononuclear cells (PBMCs) were separated from the peripheral blood by ficoll and the genomic DNA was extracted.The HIV-1 env gene C2V5 was amplified by nested-PCR and sequenced directly.The subtype A HIV-1 was identified both by sequencing and HMA.In the phylogenetic tree,the samples js01 and js03 clustered to both of the subtype A consensus and strains U455、UG06 of Uganda of the Central Africa.The genetic distance of js01 and js02 were 9.3% and 11.5% respectively from subtype A consensus.Another sample hub14 was also identified to be subtype A by HMA.The epidemiological data showed that all three samples were from persons working in Uganda,where the subtype A HIV-1 is known to be commonly found.The results above show that subtype A HIV-1 has been introduced into China by the people returned from Africa.
