摘  要:乙型肝炎病毒表面抗原-抗体复合物对抗原的加工递呈与诱生细胞因子的作用周生华闻玉梅(上海医科大学卫生部医学分子病毒学重点实验室,上海200032)关键词抗原-抗体复合物,抗原递呈,细胞因子,乙型肝炎表面抗原前曾报道,乙型肝炎(乙肝)疫苗-人抗乙肝免疫球...To study the therapeutic mechanisms of antigen-antibody complex,effects of HBsAg-mouse anti-HBs complex (IC) on HBsAg processing,presentation and cytokine induction were studied in Balb/c mice.Results showed that mouse anti-HBs and HBsAg complex constructed at the ratio of slightly excess of antigen,could significantly enhance the uptake of HBsAg by peritoneal macrophages,bone marrow-derived dendritic cells and splenocyte-derived dendritic cells.Macrophages incubated with IC efficiently potentiated the proliferation of enriched T cells from spleens of HBsAg-immunized mice.Incorporation of 3H-TdR from T cells incubated with IC-treated macrophages was 3551±525cpm,while that from T cells incubated with only HBsAg-treated macrophages was 633±32cpm,p<0.01.Splenocytes derived from IC immunized Balb/c mice produced more interferon-γ and interleukin-2,than those from only HBsAg immunized mice (IFN-γ 583.5±90 pg/ml and IL-2 365±60pg/ml in IC-immunized mice versus HBsAg-immunized mice IFN-γ 300±90pg/ml,IL-2 130±30pg/ml)
