HPLC-FLD法测定豚鼠肺组织中酯型和盐型羟基喜树碱的含量

摘　　要：目的建立豚鼠肺组织样品中羟基喜树碱酯型(C-HCPT)和盐型(0-HCPT)HPLC-FLD含量测定方法。方法以5 mg·kg^(-1)剂量给豚鼠雾化吸入羟基喜树碱注射液,于不同时间点取肺组织,样品处理后,分别测定酯型和总酯含量,减量法得到盐型的含量。色谱柱为Lichrospher C_(18)柱(250 mm×4.6 mm,5μm),C_(18)预柱(10mm×4.6mm,5μm);流动相为乙腈:0.01 mol·L^(-1)缓冲液(0.075 mol·L^(-1)醋酸胺,冰醋酸调pH至6.4)=32:68(V:V);流速为0.9 mL·min^(-1);激发波长为363 nm,发射波长为550 nm,柱温:30℃。结果豚鼠雾化吸入羟基喜树碱注射液(5 mg·kg^(-1))后,0、5、10、20 min时豚鼠肺组织中的酯型浓度分别为(8.8±s0.8)、(4.5±2.0)、(2±3)、(1±4)μg·L^(-1);盐型浓度分别(4.6±1.4)、(6.8±0.9)、(3.0±2.7)、(1±5)μg·L^(-1)。结论本法简便,专属性好,定量可靠,可满足羟基喜树碱不同构型定量研究的需要。AIM To establish a HPLC-FLD method for the determination of O-hydroxycamptothecin （O- HCPT） and C-hydroxycamptothecin （C-HCPT） in the lung of guinea pigs. METHODS After the inhalation of HCPT injection （5 mg·kg^-1） , the lung tissue samples of guinea pig were obtained at 0, 5, 10, 20, 30 min. And then ester and general ester were determined separately and the C-HCPT content was obtained by subtraction method. The concentration of HCPT was assayed on a Lichrospher C18 column with a mobile phase consisting of acetonitrile-buffer （0.075 mol·L^-1 ammonium acetate, acetate adjust pH to 6.4; 32 ： 68, V ： V） at a flow rate of 0.9 mL·min^-1, and HCPT was monitored with fluorescence detector, excitation and emission wavelengths at 363 nm and 550 nm, column temperature at 30 ℃. RESULTS When the inhalation dose was 5 mg·kg^-1, O-HCPT and C-HCPT concentration in lung of guinea pigs were （8.8 ± s 0.8）, （4.5 ± 2.0）, （2 ± 3）, （1 ± 4） μg·L^-1, （4.6 ± 1.4）, （6.8 ± 0.9）, （3.0 ± 2.7）, （1 ± 5） μg·L^-1, at 0, 5, 10, 20 min respectively. CONCLUSSION The method is simple, sensitive, reliable, and suitable for the quantitative determination of O-HCPT and C-HCPT.
