淫羊藿甙对成骨细胞凋亡的影响

摘　　要：目的通过研究淫羊藿甙对体外培养的成骨细胞凋亡的影响,探讨淫羊藿甙防治骨质疏松的可能机制。方法采用酶消化法和机械分离法获得新生SD大鼠成骨细胞,取第四代细胞随机分为,雌二醇组(1×10-8mmol/l),淫羊藿甙低剂量组(1 ng/ml),中剂量组(10 ng/ml),高剂量组(100 ng/ml)对照组,经药物处理48 h后,用TUNEL和扫描电镜的方法检测成骨细胞的凋亡。结果与对照组相比较,淫羊藿甙各剂量组的凋亡指数均下降(P<0.05),其中淫羊藿甙中剂量组的凋亡指数下降明显,差异有高度统计学意义(P<0.01);扫描电镜显示淫羊霍甙各组的凋亡细胞较对照组减少,尤以淫羊霍甙中剂量组最少。结论淫羊藿甙可以通过抑制成骨细胞的凋亡,促进骨形成,减少骨吸收,从而发挥抗骨质疏松的作用。Objective To observe the effect of icariine on apoptosis of osteoblasts in vitro and to study the possible mechanism of incariine on preventing and treating osteoporosis. Methods Calvarial osteoblasts were obtained by enzyme dissociation and mechanical isolation method from newborn SD rats, then osteoblasts of the fourth generation were divided into 5 groups randomly： 17 13-estradiol（ 1× 10^-8 mmol/l）group, low concentration of icariine group （1 ng/ml）, middle concentration of icariine group （10 ng/ml）, high concentration of icariine group （100 ng/ml） and control group. Different concentration of icariine were added into the culture mediums and apoptosis of osteoblasts were detected by TUNEL immtmohistological staining technique and electron microscope after 48 hours. Results Apoptosis indices （AI） in different concentration icariine groups were decreased as compared with that of the control group （P〈0.05）, while AI in the middle concentration icariine group （10 ng/ml） had the greatest difference （P〈 0.01 ）. There were less apoptotic osteoblasts in different icarrine groups than those in control group as studied by electron microscope scanning, especially in the middle concentration icariine group （10 ng/ml）. Conclusion Icariine can suppress apoptosis of osteoblasts to promote bone formation and decrease bone absorption. This may be one of the possible mechanisms of incariine in treating osteoporosis.
