摘 要:目的探讨中国人群内脂联素基因启动子区-11391G/A、-11377C/G基因多态性以及突变与肥胖、2型糖尿病(T2DM)的相关性。方法选取研究对象共305名,利用聚合酶链反应-限制性内切酶片段长度多态性(PCR-RFLP)方法鉴定-11391G/A、-11377C/G基因型。结果①在所选群体中未发现-11391G/A突变;②以基因型分组:-11377 GG基因型体重指数(BMI)值明显高于CC,CG基因型,且具有统计学意义(P<0.05);GG基因型舒张压、收缩压明显高于CC,CG基因型,且与CG基因型差别有统计学意义(P<0.05);CC基因型高密度脂蛋白(HDL)最高且与CG基因型差别有统计学意义(P<0.05)。结论在西安人群内未发现-11391G/A突变;-11377C/G基因多态性与BMI的相关性具有统计学意义;未发现11377C/G基因多态性与2型糖尿病的相关性。Objective To study the correlation of SNPs -11391G/A, -11377C/G with obesity and type 2 diabetes. Methods The PCR-RFLP technique was adopted to test the gene frequencies of adiponectin in 305 people. Results (1) The -11391G/A mutation was not found in our research population; O Subjects with the GG genotype at position -11377 had a significantly higher BMI than those with the GC, CC genotype (P〈0.05); more over, GG genotype at position -11377 had an increased risk of high systolic blood pressure and diastolic blood pressure compared with GC genotype (P〈0.05). HDL had a significant difference between CC and CG genotype (P〈0.05). Conclusion The-11391G/A mutation has not been found in our research population; SNP -11377C/ G may contribute to the genetic risk for BMI but not T2DM.
