Effects of Deltamethrin on Neurobehavioral Development and the Activity of Brain Na^+-K^+-ATPase in Offsprings of Intoxicated Rats

摘  要:[目的]探讨妊娠期溴氰菊酯(deltamethein,DM)暴露对仔鼠脑Na+-K+-ATPase活性及神经行为发育的影响。[方法]确定母鼠妊娠后,随机分为0、3.35及6.70mg/kgDM染毒组,自妊娠1~19d按0.5ml/100g体重隔日一次灌胃染毒,对照组给予相同剂量的色拉油。分别选择5、10、21d龄仔鼠断头处死,测其脑组织Na+-K+-ATPase活性;选择30d龄仔鼠进行主动逃避实验及旷场实验。结果]3.35mg/kg染毒组仅21d龄仔鼠存活率与对照组相比明显降低(P<0.05),6.70[mg/kg染毒组5、10、21d龄仔鼠存活率均低于对照组(P<0.01)。3.35mg/kg染毒组5d龄仔鼠脑组织Na+-K+-ATPase活性无明显变化,10d龄仔鼠活性降低,与对照组相比有显著性差异(P<0.05),21d龄仔鼠未见异常;6.70mg/kg染毒组5、10d龄仔鼠脑组织Na+-K+-ATPase活性显著降低,21d龄未见明显变化。6.70mg/kg染毒组30d龄仔鼠延迟时间为(3.86±1.40)s、被动逃避反应阳性率为24%,与对照组[(1.68±0.70)s、6%]相比,差异有显著性(P<0.05)。3.35和6.70mg/kg染毒组仔鼠旷场实验中行进格子数均明显降低,与对照组的差异均有显著性(P<0.05)。[结论]孕鼠妊娠期接触DM可以抑制仔鼠脑组织Na+-K+-ATPase活性,并对其生长发育产生影响。[ Objective ] To To study the effect of dehamethrin ( DM )on the neurobehavioral development and the activity of Na^+-K^+-ATPase of the filial brains of intoxicated rats. [ Methods ] Impregnate female rats were randomly divided into 3 groups and treated orally with 0, 3.35, and 6.70 mg/kg dehamethrin respectively every other day from pregnant day( PND ) 1 to 19. Then the activity of brain Na^+-K^+-ATPase and the neurobehavioral functions of filial rats were observed. [ Results ] The survival rate in 3.35 mg/kg group was decreased significantly at PDN 21 and that in 6.70 mg/kg group was less than the control group at either PND 5, 10 and 21 ( P 〈 0.01 ). Na^+-K^+-ATPase activity in 3.35 mg/kg group showed no change at PND 5 and 21 compared with the control group while it decresed significantly at PND 10 ( P 〈 0.05 ). This enzyme activity in 6.70 mg/kg group decreased significantly at PND 5 or 10( P 〈 0.05 )while no significant change was found at PND21. The delay time of dark escaping test in the filial rats of 6.70 mg/kg group was( 3.86 ± 1.40 )s, and the positive rate of passive escaping response was 24%, while those in control group were( 1.68 ± 0.70 )s and 6% respectively. The learning ability and memory function of filial rats in exposed group also decreased ( P 〈 0.05 ). [ Conclusions ] Exposure to high dose of deltamethrin in pregnant female rats would influence the activity of brain Na^+-K^+-ATPase and interfere the neurobehavioral development of their filial rats.
