摘 要:目的:观察外周血中糖皮质激素升高因子(GIFs)水平在GIF-HPA反馈回路及实验性自身免疫性甲状腺炎(EAT)形成中的作用,探讨AITD发病机理。方法:采用EAT易感的Lewis大鼠和非易感的Wistar大鼠,均用Tg免疫诱发EAT形成,观察免疫后不同时间(初次抗原免疫后3~4小时和8~12小时及EAT形成时),其甲状腺组织炎症反应和血清中皮质醇、GIFs(IL-1β,IL-6)和TPOAb水平。结果:诱发EAT的Lewis大鼠甲状腺炎发病率及甲状腺炎细胞浸润程度,均明显重于Wistar大鼠。Tg免疫后3~4小时,Lewis大鼠血清中皮质醇水平呈轻度升高,而Wistar大鼠明显升高。两种大鼠血清中GIFs(IL-1β,IL-6)水平动态变化与各自皮质醇水平的变化类似,免疫后Wistar大鼠血清中两种细胞因子水平的升高水平均明显高于Lewis大鼠。免疫后(3~4小时组除外)Lewis大鼠血清中TPOAb水平明显高于Wistar大鼠。结论:可以认为个体免疫应答生成的IL-1β、IL-6等细胞因子水平会影响GIF-HPA反馈回路,其异常反应也是构成AITD发病的因素。Objective:To observe the roles of the level of GIFs in GIF-HPA feedback loop and in the induction of EAT.In addition,to further understand the pathogenesis of AITD.Methods:EAT was induced by immunization with thyroglobulin in EAT susceptible(Lewis)and unsusceptible(Wistar)rats.The pathological changes in thyroid,the levels of cortisol and GIFs(IL-1β,IL-6)as well as the titers of TPOAb in the serum were determined at different times,i.e,3-4 hours after the first immunization,8-12 hours after the first immunization and two weeks after the last immunization.Results:Both the incidence of EAT and the level of infiltration of lymphocytes in thyroid of Lewis rats were much higher than those of Wistar rats.At the time of 3-4 hours after the first immunization,the levels of cortisol of Lewis rats just showed a mild increasing and no more changes could be seen at later times.Of Wistar rats,the levels of cortisol increased remarkably and even were two folds of the baseline at the time of 8-12 hours after first immunization.The pattern of the dynamic changes of the levels of GIFs(IL-1β,IL-6)in both rats was consistent with that of the changes of cortisol respectively.The titers of TPOAb in the serum of Lewis rats after immunization(excluding the point of 3-4 hours after the first immunization)were much higher than those of Wistar rats.Conclusion:Both the low production of GIFs and the discrepancy showed in the GIF-HPA feedback loop are possibly responsible for the different susceptibility of EAT of these two species of rats.Therefore,it could be considered that the level of the cytokines of IL-1β and IL-6 produced in the immune response could influence the onset of AITD via GIF-HPA feedback loop.
