摘 要:目的探索经垂直传播HIV-1env区突变准种改变HLA限制性CTL表位的方式机制。方法从2对垂直传播的HIV-1感染者血清内提取HIV-1核酸,逆转录-套式PCR(RT-nestedPCR)扩增,产物克隆后,挑选阳性克隆行CSGE分析,对优势,劣势准种克隆测序分析,对氨基酸序列中可能的CTL表位进行分析。结果第一对母子:母亲YJ体内存在HIV-1准种env区CTL表位在其54d的女儿WYF体内都存在,而WYF体内存在一株突变准种,其CTL表位在YJ体内HIV-1准种中不存在;第二对母子:母亲CM和其12mo的儿子ZDB体内HIV-1env准种无相同的CTL表位。结论HIV-1在垂直传播后,随着时间的延长,母子之间HIV-1env区突变准种所导致HLA限制性CTL表位发生改变概率增大。Objective To exploit how mutated quasispecies of HIV-1 env region change HLA restricted CTL epitodes during vertical transmission. Methods We extracted RNA from plasma of two pairs of vertical transmission individuals andpolymersed by RT-nested PCR. PCR products were T-A cloned. Positive clones were analysed by conformation sensitive gel electrophoresis(CSGE). The clones of major and minor quasispecies were sequenced. Sequencing results were anylysed for determinating CTL epitodes. Results The first pair mother-child, all CTL epitodes in env region of mother YJ's HIV-1 quasispecies existed in env region of daughter WYF's HIV-1 quasispecies, but one of CTL epitodes in env region of WYF's HIV-1 minor quasispecies was not found in env region of mother YJ's HIV-1 quasispecies; the second pair mother-child, no same CTL epitode was found in env region of mother CM's and son ZDB's HIV-1 quasispecies. Conclusion The magtitudes of HLA restricted CTL epitodes changes induced by mutated HIV-1 quasispecies during vertical transmission increase as time going.
