摘 要:目的:从变色硅胶干燥的忍冬叶片中提取高质量的基因组DNA。方法:分别采用常规CTAB法与改良CTAB法提取干叶的DNA,通过琼脂糖凝胶电泳、紫外分光光度法、酶切检测其质量,并与鲜叶的提取效果比较。结果:改良CTAB法所得DNA优于常规CTAB法,电泳条带清晰,完整性好,纯度高,能被EcoRⅠ完全酶切。从变色硅胶干燥的忍冬叶片中提取的DNA质量与鲜叶无明显差异,且产率更高。结论:改良CTAB法适合忍冬干叶基因组DNA的提取。Objective :To extract high quality genomic DNA from Lonicera Japonica Thunb. dry buds with silica gel. Methods : Two different methods ,convevtional CTAB method and modified CTAB method ,were applied to isolate genomic DNA from dry leaves of Lonicera Japonica Thunb. The DNA obtained was analyzed by the means of agarose gel electrophoresis,ultraviolet spectrophotometric method and enzymatic digestion. Results :Higher purity DNA was obtained by modified CTAB method and it can be digested by restriction enzyme EcoR Ⅰ completely. The DNA extracted from dry buds had no obvious differences with the DNA from fresh buds in quality and got higher yield.
