DMBA诱导大鼠乳腺癌癌前病变的组织病理学研究

摘　　要：目的:研究二甲基苯蒽(DMBA)诱导不同品系大鼠乳腺的病理改变,探讨造模的成模规律及成功率,建立可靠的乳腺癌癌前病变动物模型。方法:随机抽取42只SD大鼠和10只wister大鼠,分为4组:空白对照组(12只)、SD模型Ⅰ组(15只)、SD模型Ⅱ组(15只)和Wister模型组(10只),采用DMBA10mg/100g一次性灌胃造模,动态观察大鼠乳房大体形态及组织病理学改变。结果:SD模型Ⅱ组大鼠在第9周出现肉眼可见肿瘤,wis-ter大鼠未发现;空白对照组全部为正常乳腺,SD模型Ⅰ组、SD模型Ⅱ组、Wister模型组的乳腺癌癌前病变发生率分别为25%、40%、22.2%,SD模型Ⅱ组高于其他两组(P<0.05);乳腺癌发生率分别为8.3%、10%、0,SD模型Ⅱ组与SD模型Ⅰ组比较无差异,较Wister模型组发生率高(P<0.01)。结论:DMBA可诱导SD大鼠乳腺增生及癌变,造模10周后出现乳腺癌癌前病变,成功率优于wister大鼠,是癌前病变实验研究理想的动物模型。Objective ：Explore the facture of medicine - induced animal model of rat mammary cancer precancerous lesion. Methods ：52 rats were assigned to 4 groups at random： the control group were feed conventionally for 14 weeks, the SD - Ⅰ group induced SD rats with 7,12- dimethylbenz anthracene （DMBA） for 10 weeks,the SD - Ⅱ group induced SD rats with DMBA for 14 weeks, and the Wister model group induced Wister rats with DMBA for 14 weeks. After the experiment, the breast tissue was cut off and observed in Pathological Changes. Results ： All rats of the control group were normal. The incidence rate of mammary precancer of the three groups were 25% ,40% ,22.2% , and the SD - Ⅱ group was more than other two groups （ P 〈 0.05 ） ; The incidence rates of mammary cancer of the three groups were 8.3% ,10% ,0,and the SD - Ⅱ group was more than wister model groups （ P 〈0.05）. Conclusions ： Animal model of rat mammary precancer can be induced by DMBA, and mammary precancer can be established after 10 weeks. It is an ideal animal model of prevention breat cancer inhibitors.
