摘  要:目的探讨浆细胞膜糖蛋白-1(PC-1)基因K121Q多态性与2型糖尿病(T2DM)、糖尿病肾病(DN)的关系.方法应用PCR-RFLP技术检测127例T2DM患者,其中糖尿病无肾病者(NDN组)49例,DN者78例和68例健康对照者(NC组)PC-1基因K121Q的多态性分布,比较各组间基因型和等位基因分布和频率及相关临床资料.结果(1)T2DM组的KQ+QQ基因型频率和Q等位基因频率高于NC组,有统计学意义.(2)NDN组与DN组间基因型和等位基因频率无统计学差异;(3)T2DM组KQ和QQ基因型携带者的高血压发生率(63.0%)高于KK基因型(41.8%),接近差异水平(P>0.05);(4)PC-1基因K121Q多态性与DN无相关性.结论携带Q等位基因者对T2DM的易感性增强,更易患高血压.但与DN的发生、发展无相关性.Objective To investigate the relationship of K121Q polymorphism of the plasma cell glycoprotein 1 (PC-I) gene exon 4 with type 2 diabetes meUitus and diabetic nephropathy (DN). Methods The polymorphisms of PC-1 gene K121Q was detected by PCR-RFLP in 127 patients with type 2 diabetes, including 49 normal albuminuria (NDN group), 78 diabetic nephropathy (DN group) and 68 normal controls (NC group) Genotypes and allele distribution and frequencies, and its related clinical materials were compared. Results (1) The KQ+QQ genotype frequencies and Q allele frequencies in rF2DM group were significantly higher than those in NC group. (2) There were no significant differences in genotype and allele frequencies between NDN group and DN group. (3) The hypertension incidence of QQ or KQ genotype carriers (63.0%) was higher than KK genotype carriers (41.8%), which was close to statistical difference. (4) The polymorphism of PC-lgene K121Q were not correlated to DN. Conclusion The PC-1 gene exon 4 121Q polymorphism maybe risk for type 2 diabetes mellitus and hypertension development. The polymorphism of PC-1 gene K121Q may not be correlated to DN development and progression.
