摘  要:目的比较重组人巨细胞病毒(HCMV)pp65蛋白与中药金叶败毒颗粒的抗HCMV作用并探讨二者间的关系。方法用重组HCMVpp65蛋白刺激培养的人外周血单个核细胞,制备HCMV抗原特异性细胞毒T细胞,即pp65-CTL。分别将pp65-CTL、金叶败毒颗粒、pp65-CTL+金叶败毒颗粒、更昔洛韦作用于HCMV感染的人胚肺成纤维细胞(HEL),比较各组对HEL细胞病变的抑制作用,同时用RT-PCR法半定量检测作用前和作用不同时间点HCMVmRNA表达水平的变化。结果pp65-CTL组在HEL细胞感染后72h才开始出现少许细胞病变,有显著抗HCMV作用;pp65-CTL+金叶败毒颗粒组作用不同时间点对HCMVmRNA表达及HCMV所致细胞病变的抑制作用明显强于单一用药。结论重组HCMVpp65蛋白刺激培养的pp65-CTL对HCMV有明显的抑制作用,与中药金叶败毒颗粒有显著的协同作用,可共同应用于HCMV活动性感染的治疗。Objective To compare the effect of recombinant HCMV pp65 protein against HCMV with Chinese medicine Jin-Ye-Bai-Du granules and to study the relation between them. Methods The development of cytopathic effect (CPE) of human embryo lung fibroblast cell(HEL) caused by HCMV were observed in vitro before and after the treatment of pp65-CTL, Jin-Ye-Bai-Du granules, pp65-CTL + Jin-Ye-Bai-Du granules and ganciclovir, respectively. At the same time, the expression of the HCMV mRNA in infected HEL cells were measured by quantitative RT-PCIL Results There are less CPE in HCMV-in- fected HEL cells after 72h which were treated with pp65-CTL, pp65-CTL + Jin-Ye-Bai-Du granules had strongest inhibiting effect both on HCMVmRNA and CPE among five groups. Conclusion These data suggested that CTL induced by HCMV pp65 had an obvious inhibition on HCMV, and could cooperated with Jin-Ye-Bai-Du granules on action against HCMV. pp65- CTL + Jin-Ye-Bai-Du granules used as a safe and valuable medicine for treatment of active HCMV infection.
