摘  要:目的研究HIF-1α、VEGF、EphB4及其配体ephrinB2在人脑星形细胞瘤中的表达,探讨其与星形细胞瘤临床病理特征的关系。方法应用组织芯片技术及免疫组化SP法检测60例人脑星形细胞瘤和13例正常脑组织中HIF-1α、VEGF、EphB4/ephrinB2表达及微血管密度(MVD)。结果HIF-1α、VEGF、EphB4/ephrinB2蛋白在星形细胞瘤表达水平及MVDs与肿瘤临床病理分级显著相关(P<0.05),与患者年龄、性别及肿瘤的大小无关。HIF-1α、VEGF、EphB4/ephrinB2两两间均呈正相关。结论HIF-1α、VEGF、EphB4/ephrinB2过表达与肿瘤临床病理分级关系密切,可能成为判断星形细胞瘤恶性进展和预后的重要指标。Objective To study the expression of HIF-1α, VEGF and EphB4/ephrinB2 protein in human brian astrocytoma and to evaluate the relationship between the expression of the proteins and clinical pathological parameters. Method The expression of HIF-1 α,VEGF, EphB4/ephrinB2 and the MVDs in samples from 60 human astrocytoma and 13 normal brain tissues were detected by immunohistochemical and tissue chip technique. Results The expression of HIF-1 α, VEGF , EphB4/ephrinB2 and the MVDs was significantly related to tumor histopathological grade( P 〈 0.05 ), but the expression of the proteins was not related with gender, age and the tumor size. There was significant correlation between the expressions of HIF-1α. VEGF and EphB4/ephrinB2. Conclusion Overexpression ef HIF-1α, VEGF , EphB4/ephrinB2 and the MVDs is closely associated with tumor histopathological grade and might be used as one of the markers of malignant progression and poor prognosis in patients with astrocytma.
