摘  要:目的探讨嗅鞘细胞(olfactory ensheathing cells,OECs)与聚乳酸-聚羟基乙酸共聚物[poly(DL-lactide-co-gly-colide),PLGA]材料的细胞相容性。方法将新鲜分离的OECs细胞悬液接种于PLGA膜上,然后用倒置相差显微镜、扫描电镜观察细胞生长及细胞与材料的附着情况;MTT、流式细胞术等方法测定OECs的细胞增殖和细胞周期变化。结果PLGA膜上培养OECs的形态特征、增殖能力、细胞周期及倍体水平等与正常培养的OECs相比,均无显著性差异(P>0.05)。结论PLGA生物材料与OECs相容性好,适于构建组织工程化人工神经。Objective To investigate the cytocompatibility between poly (DL-lactide-co-glycolide) (PLGA) and olfactory ensheathing cells (OECs) so as to found a basis to prepare an artificial nerve made of PLGA with OECs. Methods Freshly isolated OECs were seeded on PLGA membrane, and then their growth and adhesion to the membrane were observed at different time points by phase contrast inverted microscopy and scanning electron microscopy. The proliferation and the cell cycle of OECs were examined by MTT and flow cytometry. Results There was no statistically difference between the morphological feature, proliferative capacity and cell cycle of OECs cultured on PLGA membrane and those normally cultured ( P 〉 0.05 ). Conclusion The PLGA processes good compatibility with OECs, and it is suitable to construct tissue engineering artificial nerve with OECs.
