摘 要:【目的】探讨登革病毒诱导ECV304(人脐静脉内皮细胞)细胞凋亡与胞膜死亡受体TRAILR、TNFR、Fas的表达水平的相关关系及其意义。【方法】用2型登革病毒(Dengue Virus Type2,DV2)感染ECV304细胞,流式细胞技术检测ECV304感染前、后细胞凋亡变化及死亡受体TRAILR1-4、TNFR1-2、Fas的表达水平。【结果】登革病毒感染后ECV304细胞凋亡数增加,感染前细胞凋亡数约5%(9.46%±5.07%),而感染后约28%(24.31%±3.70%),感染前、后两组细胞凋亡数经t检验,P<0.05;病毒感染后细胞表达Fas百分比增加,感染前为41%(38.95%±8.50%),而感染后为79%(67.47%±10.05%),感染前、后Fas表达经t检验,P<0.05;而TRAILR1-4,TNFR1-2表达水平甚低,感染前后无明显改变。【结论】DV2感染可诱导血管内皮细胞凋亡,其凋亡与Fas/FasL的表达改变有关,但细胞凋亡的信号转导通路有待进一步研究。[Objective] To investigate apoptosis and expression of death receptors-receptors of TRAIL, TNF, and FasL on human umbilical veno-endotheliocyte ECV304 cell strain induced by dengue virus type 2 (DV2). [Method] Flow cytometric analysis was used to detect the numbers of apoptotic cells and the expression levels of TRAILR1-4, TNFR1-2, Fas on ECV304 cells before/after DV2 infection. [Results] The numbers of apoptotic cells of ECV304 increased after DV2 infection, there were only about 5% (9.46% ±5.07%) of apoptotic cells before virus infection while there were approximately 28% (24.31% ± 3.70%) of apoptotic cells after virus infection. T test was performed in both groups, P 〈 0.05. At the same time, the expression level of Fas also increased, before virus infection about 41% (38.95% ± 8.50%) of ECV304 cells expressed Fas while 79% (67.47%± 10.05%) of ECV304 cells expressed Fas after virus infection. T test of both groups, P 〈 0.05. [Conclusion] DV2 infection can induce apoptosis of ECV304 cells, and it suggests strongly that Fas/FasL may be involved in the apoptotic signal transduction.
