摘 要:目的:观察孔雀石绿(malachite green,MG)对小鼠睾丸细胞超微结构的损伤。方法:32只雄性成年BALB/c小鼠随机均分为4组,MG浓度设为:0mg/L(对照组,A组)、100mg/L(B组)、400mg/L(C组)和800mg/L(D组)。连续灌胃给药30d,A组以等量生理盐水代替MG。应用透射电镜观察睾丸超微结构的改变。结果:B组睾丸超微结构与A组的基本相似。C组曲细精管基膜多见皱褶,支持细胞和精原细胞的胞浆空泡化及核固缩较常见。D组曲细精管基膜多呈波浪式皱褶,支持细胞和精原细胞的胞浆空泡化程度加重,核固缩明显甚至降解,管腔中多见畸形精子,间质细胞空泡化、脂滴增多。结论:MG染毒可导致大鼠睾丸曲细精管基膜、支持细胞、精原细胞、精子和间质细胞等发生超微结构的改变,剂量增加时病理改变加重。Objective: To investigate effects of malachite green (MG) on the ultrastructure of mouse testis. Methods: Thirty-two male BALB/c mice were randomly divided into four groups. MG solved in normal saline was given at the dose of 0 mg/L (group A, as control), 100 mg/L(group B), 400 mg/L(group C), or 800 mg/L(group D) for 30 d by intragastric administration consecutively. Transmission electron microscopy was used to observe changes of testicular ultrastructure. Results: The testicular ultrastructure in group B was similar with that in group A. In group C, basement membrane showed some irregular and wrinkled changes, and more vacuoles and degeneration were observed in Sertoli cells and spermatogonia. In group D, vacuoles and degeneration were common in Sertoli cells and spermatogonia. Obvious apoptosis cells and abnormal sperm heads were seen in the lumen. Leydig cells had more vacuoles and increased lipid droplets. Conclusions: MG could significantly lead to pathologic changes of ultrastructure of basement membrane, Sertoli cell, spermatogonium, sperm, and Leydig cell in mouse testis, and the injuries became more severe with the increasing doses.
