摘  要:通过对比几种标记方法,选择预亚锡化法对一种抗植物蛋白抗体进行99mTc标记,标记条件相对简单,初始标记率约为65%,经Sephadex G25凝胶柱纯化后该标记抗体的放射化学纯度达到80%以上.进一步研究了标记配合物在正常小鼠以及荷瘤(H22)小鼠体内的生物分布.生物性能研究表明:99mTc标记的抗体在小鼠体内主要经肾代谢,其在肿瘤内有一定的初始摄取,且清除较慢;靶与非靶比值随时间的增加而提高.但遗憾的是该标记配合物仍存在血液本底较高的缺点,希望通过进一步对该抗体片断的标记研究为肿瘤的早期诊断提供一种新的显像剂.A "pretinning" method was used to label antibody against a plant protein after comparison of several labeling methods. The labeling conditions were easy to achieve, with an initial labeling efficiency of 65%. After purification on Sephadex G25, radiochemical purity of the labeled antibody was 〉 80%. Biodistribution of the labeled antibody was investigated in both control and tumor-bearing mice (H22). It was found that the ^99m Tc-labeled antibody was excreted mainly by kidney, initially uptaken in tumor but cleared rather slowly. The ratios between target and non-target tissue became progressively greater with time. But the ^99mTc-labelled antibody had rather high basal blood levels. It is speculated that the labeling method could be improved to potentially provide a new method for the early diagnosis of cancer.
