实验组织切片荧光染色后再行苏木素-伊红染色的方法及体会

摘　　要：目的：初步探索荧光染色后的组织切片再行苏木素-伊红（HE）染色的技术方法和操作程序。方法：分别用5μm石蜡、冰冻两种组织切片按常规间接免疫荧光加荧光化学方法进行荧光染色,然后用pH 4.7-5.0 50%的乙醇洗脱封片的缓冲甘油,再进行常规HE染色。分别用荧光显微镜与光学显微镜观察、拍照,进行HE染色效果比较。结果：荧光染色后的组织切片标本再行HE染色,其染色效果和组织细胞结构的清晰度与组织切片单独进行HE染色相比无明显差异。结论：本实验操作方法简单;结果可靠;增加了单张切片的信息含量;节约实验成本;更为组织细胞形态学检测方法的相互转化与衔接提供了技术和思维平台。Objective： To establish a method of hematoxylin and eosin staining on the section after fluorescent labeling. Methods. Sections of paraffin and frozen ones stained with immunofluorescent or Hoechst dye were observed under a microscope. After washing away the mounting material glyencerol with 50% alcohol（ pH 4.7 - 5.0）, the slides were stained with HE. The effect of this procedure was analyzed. Results： The effect of such manipulation did not reduce the resolution. Information could be obtained both from light and fluorescence microscopes. Conduslon： We have introduced a new method to broaden the morphological information from one section without torturing the morphological structure.
