摘  要:目的探讨促血小板生成素(TPO)对小鼠骨髓有核细胞(BMNc)的细胞周期及骨髓干细胞增殖的影响。方法将昆明小鼠随机分为实验组与对照组,每组15只。实验组腹腔注射TPO 50μg·kg^-1·d^-1,连用5d;对照组腹腔注射生理盐水0.1ml·g^-1·d^-1。每组分别于最后1次注射后6h、12h及168h收集BMNC,检测BMNC的细胞周期及表达CD34’的抗原绝对数;计数BMNC,以1×10^6细胞/cm^2接种培养并计数原代成纤维样细胞集落形成单位(CFU-F)。结果应用TPO后的细胞周期显示,实验组在6h和12h取材BMNC的G0/G1期细胞百分率较对照组低12%~8%(P〈0.05),而S+Q期增高12%~8%(P〈0.05);实验组BMNC表达CD34^+的抗原绝对数较对照组明显增加1~2倍(P〈0.05);实验组所获得CFU-F集落数比对照组增加3~9倍(P〈0.01)。结论TPO促进CD34^+细胞增加的同时,还可促进小鼠BMNC进入细胞周期,促进CFU—F集落数增多,即促进骨髓间充质干细胞(MSC)增殖,有利于改善造血干细胞移植(HSCT)的效果。Objective To investigate the effect of thrombopoietin (TPO) on the cell cycle of bone marrow nucleated cells (BMNC) and the proliferation of bone marrow stem cells in mice. Methods Kunming mice were divided into TPO-treated and control groups randomly (n = 15 in each group) and subjected to intraperitoneal injection of TPO at a dose of 50 μg/kg and saline at a dose of 0. 1 ml/g per day for 5 days respectively. The BMNC were harvested at 6, 12 and 168 h after the final administration. The cell cycle and CD34 antigen expression in BMNC were analyzed by flow cytometry. BMNC were counted and seeded at a density of 106 cells per cm2. The colony-forming unit-fibroblast (CFU-F) of P0-MSC was cultured and evaluated. Results Compared with control group, the percentage of BMNC, harvested at 6 and 12 h, in G0/G1 phases was decreased (12%-8 %) (P〈0. 05), while that in S + G2/M phases increased ( 12%-8 %) in TPO group (P〈0. 05). The CD34^+ cell number in TPO group was increased by 1-2 times as compared with that in control group (P〈0. 05). The number of CFU-F in TPO group was also increased by 3-9 times as compared with the in control group (P〈0. 01). Conclusion After administration of TPO, the cell number of CD34^+ was increased, BMNC could enter into cell cycles and proliferation of MSCs could be promoted, which was beneficial to the improvement of HSCT effects.
