RP-HPLC测定猪胆粉薄荷脑软膏中猪去氧胆酸的含量

摘　　要：目的：建立猪胆粉薄荷脑软膏中猪去氧胆酸的含量测定方法。方法：采用RP-HPLC法，以ZorbaxSBC18为色谱柱，流动相为甲醇-0．1％甲酸溶液（80：20），流速为0．5mL·min^-1，采用蒸发光散射检测器，漂移管温度为90℃，氮气流速为1．6L·min^-1。结果：猪去氧胆酸线性范围为0．7068～10．602μg，r=0．9993。猪去氧胆酸的平均回收率为97．61％，RSD为2．4％。结论：本方法定量准确，重复性好，可用于猪胆粉薄荷脑软膏的质量控制。Objective：To establish the method for determining the content of Hyodeoxycholic Acid in Porcin Gallbladder Powder and Menthol Ointment by RP-HPLC. Methods：The Zorbax SB C18 column with methanol -0. 1% formic acid（80：20） as the mobile phase was used. The flow rate was 0. 5 mL· min ^-1 and the ELSD was used, the tube temperature was 90℃ and the flow of nitrogen is 1.6 L · min ^- 1. Results： The calibration curve was linear at a range of 0. 706 8 - 10. 602 μg for the hyodeoxycholic acid （ r = 0. 999 3 ）. The average recovery was 97. 61% and the relative standard deviation （RSD） was 2. 4% （ n = 9）. Conclusion： The method is accurate with a good reproducibility and can be used as a quantitative analysis method for this preparation.
