摘 要:目的探讨骨癌痛大鼠背根神经节(DRG)电压依赖性钠通道Nav 1.8 mRNA的表达。方法成年雌性SD大鼠16只,体重180—200g,随机分为对照组和骨癌痛组,每组8只。对照组腹腔注射10%水合氯醛300mg/kg麻醉后处死,冰上取L5.6 DRG。骨癌痛组采用胫骨内注射肿瘤细胞的方法制备大鼠骨癌痛模型,术前及术后14d测定大鼠机械痛阈和CO2激光热痛阈,术后14d处死大鼠,取L5.6 DRG。采用定量PCR方法测定DRG Nav 1.8 mRNA的表达。结果骨癌痛组机械痛阈与热痛阈较术前明显降低(P〈0.01);与对照组相比,骨癌痛组术后14d DRG Nav 1.8 mRNA表达上调(p〈0.01)。结论骨癌痛大鼠DRG Nav 1.8 mRNA表达上调,该通道可能参与骨癌痛的发生。Objective To investigate the expression of voltage-gated sodium channel Nav 1.8 in dorsal root ganglion (DRG) in a rat model of bone cancer pain. Methods Sixteen adult female SD rats weighing 180-200 g were randomized into 2 groups ( n = 8 each) ; Ⅰ control group and Ⅱ bone cancer pain group. The animals were anesthetized with intraperitoneal 10% chloral hydrate 300 mg/kg and killed and the L5.6 DRGs were removed in the control group. The rats in bone cancer pain group received intra-tibial inoculation of Walker 256 mammary gland carcinoma cells. Threshold to yon Frey filament stimulation and noxious thermal stimuli were measured before and 14 days after inoculation of carcinoma cells. The L5,6 DRGs were removed 14 days after carcinoma cell inoculation. The expression of Nav 1.8 mRNA in DRGs was detected by quantitative PCR. Results The animals in cancer pain group displayed Significant decrease in the threshold to van Frey filament and noxious thermal stimulation at 14 day after inoculation as compared with the baseline values before inoculation. The expression of Nav 1.8 mRNA in DRG was significantly up-regulated in bone cancer pain group as compared with the control group. Conclusion Bone cancer pain results in up-regulatlon of the expression of Nav 1.8 mRNA in DRG suggesting that Nav 1.8 may be involved in inducing bone cancer pain.
