益气化瘀中药对大鼠胃癌前病变蛋白激酶C及细胞周期素E蛋白表达的影响

摘　　要：目的观察益气化瘀中药对大鼠胃癌前病变胃黏膜蛋白激酶C(PKC)、细胞周期素E(CyclinE)表达的影响。方法采用甲硝基亚硝基胍(MNNG)、乙醇、饥饱失常等多因素造模。造模成功后,将造模存活大鼠随机分为自然恢复组及中药高、中、低剂量组,各组大鼠做相应的处理后,免疫组化法检测各组大鼠胃黏膜组织PKC、CyclinE表达水平。结果各治疗组PKC表达及CyclinE蛋白阳性率较自然恢复组下降,有显著差异(P<0.05)。结论益气化瘀中药可能是通过下调PKC及CylinE的表达,抑制细胞增殖,诱导细胞凋亡,而发挥对大鼠胃癌前病变的治疗作用。Objective To observe the effect of Yiqi Huayu Recipe on the expression of protein kinase C （PKC） and CyclinE in rats with gastric precancerous lesions （GPL）. Methods The model of GPL was established mainly through N-methyl-N-nitro N-nitrosoguanidine （MNNG） together with other factors including alcohol stimulation and undue hunger and overeating. The survived rats after modeling were randomly divided into spontaneous recovery, and high-, moderate- and low-dose Yiqi Huayu group. After the corresponding treatment, the expression of PKC and CyclinE were detected by immunohistochemical method in each group. Results The expression of PKC and CyclinE in trement group were distinctly lower than those in spontaneous recovery group （P〈0.05）. Conclusion Yiqi Huayu Recipe was effective in the treatment of rats with GPL by inhibiting the expression of PKC and CyclinE.
