摘  要:目的:观察川芎嗪对幼鼠反复惊厥后海马丙二醛(malondialdehyde,MDA)含量、超氧化物歧化酶(super oxidedis-mutase,SOD)活性、总抗氧化能力(total antioxidative capability,T-AOC)及脑含水量变化的影响。方法:162只20日龄健康Sprague-Dawley(SD)大鼠随机分为3组:对照组、惊厥组及川芎嗪干预组。通过三氟乙醚反复吸入(连续6次,qd)制作发育期大鼠惊厥动物模型。取海马组织匀浆,检测各组动物反复惊厥后6h,1,3,7d海马组织MDA含量,SOD活性、T-AOC水平变化,同时观察脑含水量变化和光镜下海马区神经元病理改变,并对脑损伤进行神经病理半定量积分。结果:川芎嗪干预组各时间点MDA含量均较惊厥组显著降低,SOD活性及T-AOC水平显著增高(P<0.01);脑含水量及脑损伤病理积分显著降低(P<0.01);海马神经元水肿、变性坏死明显减轻。结论:川芎嗪对幼鼠惊厥性脑损的保护机制可能与提高海马SOD活性及T-AOC水平、降低MDA含量有关。OBJECTIVE To investitage the effect of tetramethylpyrazine on malondialdehyde (MDA), super oxidedismutase (SOD) and total antioxidative capability (T-AOC) in the hippocampus and water content of brain tissues of the developing rats following recurrent seizures. METHODS 162 postnatal Sprague-Dawley rats of 20-day old were randomly divided into three groups: the control group, the seizure group and the tetramethylpyrazine treated group. Seizure model of rats were induced by inhalant flurothyl daily in 6 consecutive days. Brain tissue was sampled after 6 hours, 1 day,3 days and 7days in each group after last seizure,and the content of MDA,SOD activity and the level of T-AOC were detected. At the same time, the water content was detected and the pathological changes in the hippocampus of rats were observed after recurrent seizures and the brain injury was evaluated by using a semiquantitative neuropathological scoring system. RESULTS In tetramethylpyrazine treated group, the activity of SOD and the level of T-AOC in the hippocampus at different time points were much higher than those in seizure group(P〈0. 01 ), while the content of MDA, water content of brain tissues and europathological score were obviously lower than those in seizure group(P〈0. 01 ) ; edema,degeneration and necrosis of nerve cells in the hippocampus of the rat were also significantly decreased. CONCLUSION The protective effect of tetramethylpyrazine on the developing brain injury induced by seizures is probably related to raising of activity of SOD and the level of T-AOC,and decreasing of the content of MDA in the hippocampus.
