摘  要:目的观察亚硒酸钠(Na2SeO3)与氯化锰(MnCl2)单独及联合作用对二氧化硅(SiO2)致肺泡巨噬细胞(AM)超氧化物歧化酶(SOD)、丙二醛(MDA)、过氧化氢(H2O2)及过氧化氢酶(CAT)水平变化的影响,寻找两者的最佳剂量组合。方法采用大鼠肺灌洗法纯化获得AM(1×1012个/L),同时加入SiO2及不同浓度的Na2SeO3溶液、MnCl2溶液、Na2SeO3+MnCl2溶液,37℃,5%CO2培养箱培养18h后,检测MDA、H2O2含量、SOD及CAT活力。结果Na2SeO3与MnCl2单独及两者联合使用均具有拮抗SiO2细胞毒性作用。且以1.0μmol/LNa2SeO3与1.00mg/LMnCl2联合作用效果最好。结论Na2SeO3与MnCl2单独及两者联合应用在体外可有效拮抗SiO2粉尘的细胞毒性作用,降低AM的MDA、H2O2含量,同时升高SOD及CAT活力,两者合用效果更佳。Objective To investigate the effect of sodium selenite and manganese chloride used either solely or together in antagonism of the toxicity of silica on alveolar macrophages. Methods Alveolar macrophages were cultured with silica. At the same time sodium selenite, manganese chloride and manganese chloride together with Sodium selenite were added respectively to the culture. Malondialdehyde (MDA), superoxidedismutase ( SOD), hydrogen peroxide ( H2O2 ) and catalase (CAT) in alveolar macrophage were measured. Results Sodium selenite, manganese chloride, manganese chloride together with sodium selenite, had effects to contradict the toxicity of silica on alveolar macrophages. The most suitable combination of the two substances should be 1.00 mg/L of manganese chloride with 1.0 μmol/L of sodium selenite. Conclusion Sodium selenite and manganese chloride, used both solely and together could reduce the levels of MDA and H2O2 ,while increase the activities of SOD and CAT in alveolar macrophages exposed to silica.
