摘  要:目的研究运动训练对大鼠脑梗死后缺血半暗带组织中突触结构的影响,探讨神经功能可塑性的机制。方法选用SD雄性成年大鼠制成右侧大脑中动脉梗死模型56只,于造模后24h随机抽取8只作取材定位组,其余48只5d后随机分为对照组和训练组,每组24只,2组再分10,20,30d组,每组8只。训练组给予运动训练,对照组仅在笼中安静饲养。观察运动训练对大鼠学习记忆能力及缺血半暗带脑组织突触结构参数的影响。结果电镜下观察到各期训练组缺血半暗带组织突触后致密物厚度、活性区宽度、突触后膜曲率和穿孔性突触百分率均较对照组明显增加,20,30d组改变更明显(P〈0.01)。相应行为学检测训练组的20,30d组在Y迷宫分辨学习和一次性被动回避反应测试中记忆力明显优于对照组(P〈0.05)。结论运动训练可明显促进缺血半暗带脑组织突触结构的变化,以提高突触传递效率,增强记忆力。Objective To study the effects of motor training on synaptic structures in ischemic penumbra and to evaluate the relation between learning and memory ability and the change of synaptic structures. Methods Fiftysix male adult Sprague-Dawley rats were used to establish the models of right middle cerebral artery occlusion, from which, 8 rats were randomly selected and served as allocation group after 24 hours, the remaining 48 rats were divid- ed randomly into a training group and a control group after 5 days of the establishment of models, and further divided into 10-,20-,30-day subgroups. All the rats in the training group were intervened with motor training, while those in the control groups were reared in their original living state. Synaptic ultrastructure in ischemic penumbra was observed morphologically and quantitatively, and the learning and memory ability were assessed at the same time. Results It was shown that, when compared with those of the control group, the synaptic curvatures and post-synaptic density (PSD) as well as the percentage of perforated synapses were significantly increased in the rats of training group ( P 〈 0.01 ), and the learning and memory abilities were significantly improved in the rats following rehabilitation motor training (P 〈0.05). Conclusion It is concluded that motor training can facilitate the the changes of synaptic ultrastructure in the ischemic penumbra, which might contributed to the improved learning and memory ability.
