摘  要:目的探讨地塞米松对大鼠晶状体上皮细胞TGF-β受体表达的影响及意义。方法取1月鼠龄SD大鼠,予地塞米松眼膏涂双眼结膜囊,分别于用药1月、3月及6月摘取晶状体作为实验材料,采用晶状体前囊膜铺片方式,应用荧光免疫组化技术进行TGF-βR1和TGF-βR2荧光免疫组化染色,检测其蛋白水平表达的变化;采用剥取晶状体囊膜组织方法,应用RT-PCR技术检测TGF-βR1和TGF-βR2在转录水平表达的变化。取相同数量的正常同龄大鼠作为对照组。结果在蛋白及转录水平,TGF-βR1和TGF-βR2的表达于用药后1、3、6月均较同龄对照组减弱(P<0.05);对照组中,TGF-βR1和TGF-βR2的表达随着年龄的增加也逐渐减弱(P<0.05);实验组中,随着用药时间的增加,TGF-βR1和TGF-βR2的表达进一步减弱(P<0.05)。结论地塞米松可以降低大鼠晶状体上皮细胞中TGF-βR1和TGF-βR2的表达,并随着用药时间的延长,其表达进一步降低,对白内障的形成有一定的影响。Objective To investigate the effect of dexamethasone on the expressions of TGF-β receptors in rat lens epithelial cells. Methods Sprague Dawley rats aged 1 month old were used. Lenses were extracted from freshly obtained enucleated rat eyes respectively after 1-, 3-, and 6-month topical application of dexamethasone ointment to conjunctiva. The protein and mRNA expressions of TGF βR1 and TGF-βR2 in rat lens epithelial cells were detected by immunofluorescence technique and RT-PCR respectively. The rats matched with the experimental rats were used as controls. Results TGF-βR1 and TGF-βR2 expressions at protein and rnRNA levels after 1- , 3-, and 6-month application of dexamethasone were lower than those of controls (P〈(0. 05) and those in controls showed a significant age-related decline (P〈0. 05). TGF-βR1 and TGF-βP,2 protein and rnRNA expressions in experimental rats decreased with the prolonged time of treatment (P〈0. 05). Conclusion Dexamethasone decreases TGF-βR1 and TGF-βP,2 expression at protein and rnRNA levels in rat lens epithelial cells in a time-dependent manner, indicating its possible role in preventing cataract.
