摘 要:目的探讨纳米羟基磷灰石/聚酰胺66(Nano—hydroxyapatite/Polyamide66,n—HA/PA66)松质骨螺钉的生物活性及其固定山羊股骨髁骨折的可行性。方法选取成年山羊30只,随机分成两组,建立股骨外髁骨折模型。实验组用2枚n—HA/PA66螺钉固定,对照组用2枚国产聚-DL-乳酸(Poly-DL-lactide,PDLLA)螺钉固定,术后外固定6周,于术后4,8,12周摄x线片、行组织学以及生物力学检测,即刻固定强度行体外实验。结果X线片显示,骨折在8周时均骨性愈合,骨折未见移位或畸形愈合。组织学切片显示,4周时n—HA/PA66螺钉与骨界面上有新生骨痂样组织形成;8周时n—HA/PA66螺钉周围新生骨组织形态类似编织骨;12周时n—HA/PA66螺钉周围形成成熟骨小梁。结论n—HA/PA66螺钉具有成骨活性,最终与骨组织发生整合,不需再次手术取出,有望成为一种新的内固定材料。Objective To investigate bioactivity of the Nano-hydroxyapatite/polyamide66 (n-HA/ PA66) cancellous screw and its feasibility in fixation of femur condyle fracture in adult goats. Methods A total of 30 adult goats were selected and divided into two groups randomly to establish the animal models of lateral of femur condyle fracture. The experimental group was fixed with 2 n-HA/PA66 cancellous screws and the control group with 2 Poly-DL-lactide (PDLLA) cancellous screws made in China. Each goat was fixated externally for 6 weeks after operation. X-ray examination, histological examination and biomechanical test were done to observe the fixation strength at 4, 8,12 weeks after operation. The instant fixation strength was tested in vitro. Results The imageology examination showed bony healing of the femur condyle fractures at the 8th week, without displacement or malunion of fracture. Histological tests indicated new regeneration fibrous callous on the interface at 4 weeks, new regeneration bone demineralized well on the interface at 8 weeks and mature trabecula of bone around the screw at 12 weeks after operation in n-HA/PA66 group. Conclusion The nano-hydroxyapatite/polyamide66 screw has bioactivity of bone formation, can be integrated with bone tissues and is hopeful to become a new internal fixation material.
