摘 要:目的通过测定体外培养的SD大鼠胃肠平滑肌细胞内游离Ca2+浓度,观察胃肠舒对SD大鼠胃肠平滑肌细胞内游离Ca2+浓度的影响,探讨胃肠舒促胃肠动力的作用机制。方法离体培养SD大鼠胃肠平滑肌细胞,应用特异性Ca2+荧光探针Fura-2AM负载细胞,激光共聚焦显微镜检测游离Ca2+浓度。结果胃肠舒在25、50、100 mg/ml时均可升高胃肠平滑肌游离Ca2+浓度,且随剂量增加而加强。结论胃肠舒具有升高胃肠平滑肌细胞游离Ca2+的作用,Ca2+作为细胞内第二信使可能参与了胃肠舒促进胃肠平滑肌细胞的动力机制。Objective To observe Weichangshu on free Ca^2+ concentration in gastrointestinal smooth muscle cell and explore its mechanism through determining free [ Ca^2+ ] i concentration in rat gastrointestinal smooth muscle cell. Methods SD rat gastrointestinal smooth muscle cells were cultured and loaded with Fura-2AM. [Ca^2+]i was measured by fluorescent intensity in each group cell with laser scanning confocal microscope. Results 25, 50, 100 mg/ml Weichangshu could elevate [ Ca^2+ ] i in gastrointestinal smooth muscle. The level of [Ca^2+]i acted in dose-dependent. Conclusion The data indicated Weichangshu could increase [Ca^2+ ]i in gastrointestinal smooth muscle cells. Ca^2+ as an intracellular second messenger, might be involved in the mechanism by which Weichangshu promoted the motility of gastrointestinal smooth muscle cells.
