摘  要:目的研究蛋氨酸和维生素C联合拮抗铅对大鼠海马组织一氧化氮合酶(NOS)活力、一氧化氮(NO)水平和学习记忆的影响。方法SD乳鼠(85.4±9.1 g)22只,雌雄各半,随机分为实验组10只,对照组12只。各组SD大鼠饮用染铅水(硝酸铅和去离子水按1.3 g/L配成),同时实验组大鼠用蛋氨酸加维生素C每周5次,共8周,对照组仅用去离子水灌胃。测定大鼠血铅值、海马组织NOS活力和NO水平,并用Morris水迷宫实验进行学习记忆能力的测试。结果给予蛋氨酸和维生素C组大鼠的血铅值(0.482±0.057μmol/L)明显低于对照组(血铅值:3.44±0.031μmol/L),而海马组织NOS活力(41.91±7.77μmol/L)和NO水平(25.30±14.20μmol/L)明显高于对照组(NOS 6.61±3.01,NO 11.80±8.95μmol/L),t检验均具差异有统计学意义。海马组织的NOS活力、NO水平与血铅值成负相关性(r=-0.955,P<0.01;-0.490,P<0.05),Morris水迷宫第四天成绩与血铅值成呈正相关(R=0.497,P<0.05),而且实验组潜伏期短于对照组(t=-2.180,P<0.05)。结论蛋氨酸、维生素C可通过降低血铅水平,提高铅染毒大鼠海马组织的NOS活力及NO水平,而改善染铅大鼠的学习记忆功能。Objective To study the combinational effect of methioine and vitamin C on nitric oxide synthase(NOS) activity and nitric oxide(NO) level in brain of SD rats induced by lead. Methods Measurement of NOS activity and the level of NO in brain and Morris water maze test were adopted. Experimental group SD rats were fed to the concentration of lead nitrate (1.3 g/L) in water solution, and were administered two nutrients (methioine and vitamin C) compared with control group of the same concentration lead solution but only were administered water. Results The NOS activity and the level of NO in brain were increased( P 〈 0.01, P 〈 0.05), and the level of lead in the blood was decreased in the rats fed methioine and vitamin C( P 〈 0.01 ). NOS, NO and the level of bead in the blood were opposite relation( r = - 0.955, P 〈 0.01 ; r = - 0.490, P 〈 0.05) ,The escape latency in Morris maze on day 4 and the level of bead in the blood were relation( r = 0.497, P 〈 0.05), and experimental group were less than in control group( P 〈 0.05). Conclusion Methioine and vitamin C might reduce body lead and increase NOS and NO, and improve learning and memory of SD rats induced by lead.
