载脂蛋白E基因多态性与冠心病关系的研究

摘　　要：目的：探讨载脂蛋白（Ａｐｏ）Ｅ基因多态性与冠心病的相关性，以及ＡｐｏＥ基因多态性对冠心病患者血脂水平的影响。方法：１００例冠心病患者和４３例正常对照者。按常规方法测定血浆脂质和Ａｐｏ水平。ＡｐｏＥ基因型的确定采用聚合酶链反应和ＨｈａＩ酶切的方法。结果：本研究只发现３种常见的ＡｐｏＥ基因型，即Ｅ３／３，Ｅ３／４，Ｅ３／２。在病例组和正常对照组之间，ＡｐｏＥ等位基因频率以及基因型频率分布没有统计学差异（Ｐ＞０．０５）。在冠心病患者不同基因型之间，总胆固醇和ＡｐｏＢ的水平有差异（Ｐ＜０．０５）；其它血脂指标无差异（Ｐ＞０．０５）。结论：本研究证实ＡｐｏＥ基因多态性影响冠心病患者总胆固醇和ＡｐｏＢ的水平；但ＡｐｏＥ基因多态性并不是冠心病发病的直接危险因子。Objective:To elucidate the relationship between apolipoprotein(Apo)E gene polymorphism and coronary heart disease(CHD) and to evaluate the effect of Apo E gene polymorphism on the lipid levels of CHD patients. Methods:One hundred CHD patients and 43 normal controls were included.Serum lipid levels were measured according to routine methods.Polymerase chain reaction(PCR),HhaI digestion,polyacrylamide gel electrophoresis,and silver staining were used for the detection of Apo E genotype. Results:Only 3 common genotypes of Apo E, E3/3,E3/4 and E3/2 were detected in the study.There were no statistically significant differences in Apo E allele or genotype frequencies between CHD patients and controls ( p >0 05).The genotypes were associated with serum total cholesterol(TC) and Apo B levels ( p <0 05).No association was found between Apo E genotype and other lipid variables( p >0 05). Conclusion:The study has confirmed that Apo E gene polymorphism affects serum TC and Apo B levels of CHD patients,but it is not a direct risk factor of CHD.
