摘 要:本实验建立了一种浮动基点式 fura—2测定离体灌流心脏细胞内游离钙浓度的新模型。本模型中浮动基点式测定法克服了目前 fura—2技术中受细胞内某些物质产生的自身荧光变化干扰的缺点,实现了自动消除细胞自身荧光变化对实验结果的影响,并在此基础上将广泛用于测定游离状态的细胞内游离钙的 fura—2技术推广到了离体灌流心脏,实现了在更接近心肌生理状态的条件下测定其胞浆游离钙。A model of measuring cytosolic free calcium concentration of isolated perfusion hearts with fura-2 fluorescence technology was established on a self-made apparatus.A floating fluorescence base point record method was used in the model.The new recrd methd can autmatically cancel the effect of cell-autofluorescence change on experiment results at any time,therefore solved effectively the interference from the autofluorescence.On the basis Of the new record method,fura-2 fluorescence technology was popularized to isolated perfusion hearts from cell-free state.On the model,myocardium cytosolic free calcium was measured on the condition that myocytes were better cIose to their physiological state.
