摘  要:目的探讨面神经损伤动物模型中CD^+4T细胞与损伤的面运动神经元(facialmotoneuron,FMN)之间的关系,为揭示面神经损伤后修复及再生中的免疫病理机制提供实验证据。方法磁珠分选法分离野生型BALB/c小鼠CD^+4T细胞,经尾静脉移植于相同遗传背景的裸小鼠,并以PBS为对照。1周后均制备成周围性面神经损伤模型,指定时间在面神经断端涂抹荧光金示踪剂。术后3d灌注固定动物,收集脑干切片,荧光显微镜下观察。采用Image Pro Plus5.1图像分析软件计数FMN,并比较不同组之间的差异。结果术后3dCD^+4T细胞移植组FMN计数为3444.5±84.2(x±s,下同),对照组FMN计数为3013.2±65.3,差异具有统计学意义(t=5.52,P=0.0003);而CD^+4T细胞移植组与野生型小鼠神经元计数比较,差异无统计学意义(t=0.49,P=0.6347)。结论CD^+4T细胞可以保护裸鼠受损伤的FMN,使其数量达到野生型的水平。Objective To investigate the specific T cell subpopulation and the relationship with facial motoneuron in immune deficiency mouse model with facial nerve paralysis, so as to find information for new strategy of facial palsy treatment. Methods Firstly, purifying the CD^+4 T cell from wild type mouse and reestablishing the immune function of nude mouse by infusing the CD^+4 T cell through the tail vein a week before the surgery. Then the all nude mouse (BALB/c background) and wild type mouse (BALB/c background) were subjected to a right facial nerve axotomy. Then the mouse was studied by application and assessment with fluorogold retrotracer at specific time. After collecting the slices of brain stem three days post the operation, the facial motoneurons was observed under fluorescence microscope, then analyzed and counted with the software Image Pro PlusS. 1. Results The number of survival facial motoneuron in the group with CD^+4 T cell transplantation and control group was ( 3444. 5 ± 84. 2 ,x ± s) and ( 3013.2 ± 65. 3 ) respectively. There was significant difference of the number of survival facial motoneurons between nude mouse transplanted with CD^+4 T cell and PBS at three days post the operation ( t = 5.52, P = 0. 0003 ). But there was no significant difference of survival facial motoneurons between nude mouse transplanted with CD^+4 T cell and wild type mouse three days post the operation (t = 0. 49, P = 0. 6347 ). It was the transplantation of CD^+4 T cell that rescued the survival facial motoneuron to the level of wild type. Conclusions CD^+4 T cell have the ability to rescue the injuring facial motoneuron from death. It may suggest that there is a critical role of the specific T cell subpopulation in facial nerve repair and regeneration.
