摘  要:目的研究一种从鲨鱼软骨简便、快速提取低分子蛋白的方法,并证实该方法提取的鲨鱼软骨低分子蛋白能有效的抑制新生血管的形成。方法以鲨鱼软骨为原料,粉碎后反复冻融,离心去沉淀,超滤、真空干燥后得到鲨鱼软骨低分子蛋白。体外实验测定其对内皮细胞增殖、黏附和迁移的影响,用鸡胚绒毛尿囊膜实验测定对新生血管形成的影响。结果鲨鱼软骨低分子蛋白能显著抑制血管内皮细胞的生长抑制率为(36.77±2.98)%、黏附抑制率为(52.95±4.48)%和迁移运动抑制率为(68.05±4.04)%,可显著抑制鸡胚绒毛尿囊膜的新生血管的形成。结论本研究所提取的鲨鱼软骨低分子蛋白可以抑制内皮细胞生长、黏附和迁移运动,进而抑制新生血管的生长。Objective To find a way to abstract the low molecular weight protein from shark cartilage simply and quickly,and confirm the protein can inhibit angiogenesis growth. Methods First ,we got the protein by dealing with the shark cartilage through crashing, repeated freezing-thawing, centrifuging to remove the sediment, ultrafihrating and vacuum drying in order. Second, we observed the influence of the protein on the growth of endothelial cell in vitro and determined the influence on angiogenesis of embryonated egg's allantocherion, Results The protein could inhibit significantly vascular endothelial cell's growth (inhibitory ratio: 77% ± 2.98% ), adhesion (inhibitory ratio :52.95% ± 4.48% ) and locomotory movement (inhibitory ratio :68.05% ± 4.04% ). Further,it inhibited the formation of angiogeneais of embryonated egg's allantocherion. Conclusion The protein abstracted from shark cartilage could inhibit the growth of angiogenesis by inhibiting the growth ,adhesion and locomotory movement of endothelial cell.
